Yes, you have to go through immigration. There are no rules or restrictions, so it is fine to do a visa run in one day.
The good thing about Singapore is that it is possible to do a visa run in one day. It is cheaper however to fly to Kuala Lumpur, as Air Asia fly to KL and you can get flights at rock bottom prices. However, because of timings, you have to fly to KL, stay overnight (in the airport if you like) and fly back the next morning.
