US notes for arrival question?

Apologies if this sounds totally blonde... When I go to the PO to get american dollars do I ask for 6 x US$25 or can I just get US$150 as it comes even if its a hundred dollar bill? And we can hand it over altogether at the airport? Thanks for your help.
