Of course, it can be done in one day, but why not "kill two birds with one stone" and make it a 2-day trip?
First, avoid the southern road going to Gilimanuk, with all its traffic of large trucks, buses, etc. Cut across instead through Bedugul and Munduk: it will be as long (from Kuta about 3 hrs) and maybe a little less. There are some homestays in Gilimanuk, but you can stay o/n in one of the hotels in/near the National Park, or, better yet, in Pemuteran (about 20 - 25 min. from Gilimanuk). Beautiful accommodations, nice beach, excellent snorkelling and diving.
